Clozapine is an atypical antipsychotic drug, distinguishing itself from traditional antipsychotics by its unique interaction with dopamine receptors. It affects dopamine-mediated behaviors differently compared to other typical antipsychotic drugs. Clozapine binds to D1, D2, D3, and D5 dopamine receptors, with a particularly strong affinity for the D4 receptor. This pattern of activity is thought to primarily affect limbic dopamine receptors, which may explain Clozapine’s minimal extrapyramidal side effects. Additionally, Clozapine acts as an antagonist at adrenergic, cholinergic, histaminergic, and serotonergic receptors.
